Prestinhasbeenidentifiedasamotorproteinresponsibleforouterhaircell(OHC)electromotilityandisexpressedontheOHCsurface.PreviousstudiesrevealedthatOHCelectromotilityanditsassociatednonlinearcapacitanceweremainlylocatedattheOHClateralwallandabsentattheapicalcuticularplateandthebasalnucleusregion.ImmunofluorescentstainingforprestinalsofailedtodemonstrateprestinexpressionattheOHCbasalendsinwhole-mountpreparationoftheorganofCorti.However,therelacksadefinitivedemonstrationofthepatternofprestindistribution.TheOHClateralwallhasatrilaminateorganizationandiscomposedoftheplasmamembrane,corticallattice,andsubsurfacecisternae.Inthisstudy,thelocationofprestinproteinsindissociatedOHCswasexaminedusingimmunofluorescentstainingandconfocalmicroscopy.Wefoundthatprestinwasuniformlyexpressedonthebasolateralsurface,includingthebasalpole.Nostainingwasseenonthecuticularplateandstereocilia.Whenco-stainedwithamembranemarkerdi-8-ANEPPS,prestin-labelingwasfoundtobeintheouterlayeroftheOHClateralwall.Afterseparatingtheplasmamembranefromtheunderlyingsubsurfacecisternaeusingahypotonicextracellularsolution,prestin-labelingwasfoundtobeintheplasmamembrane,notthesubsurfacecisternae.ThedatashowthatprestinisexpressedintheplasmamembraneontheentireOHCbasolateralsurface.
