简介：CD134,amemberofthetumornecrosisfactorreceptorsuperfamily,playsacrucialroleinTcellsurvival.Inthisstudy,peripheralbloodmononuclearcells(PBMCs)stimulatedwithphytohemagglutinin(PHA,50μg/ml)weretreatedbyCD134mAb(1μg/ml,5μg/ml,10μg/ml)for6h,12h,24hand48h.ThelevelofperforinmRNAwasmeasuredbyreversetranscription-polymerasechainreaction(RT-PCR)technique.OurdatashowedthattheexpressionofperforinmRNAinPBMCswasdown-regulatedbyCD134mAbinadose-dependentmannerinrangeof1μg/mlto5μg/mlanddroppeddowntoitsminimumon24h(p＜0.05).ThelevelofperforinmRNAreachedaplateauwhentheconcentrationofCD134mAbexceeded5μg/ml.Inconclusion,CD134mAbcaninhibitperforinexpression,whichmayenhancetheabilityofTcellsforsurvival.
