  • 简介:AbstractOxidative stress is caused by the imbalance between the generation of free radicals/reactive oxygen species (ROS) and the antioxidant defense systems, which can activate various transcription factors and affect their transcriptional pathways. Oxidative stress plays an important role in the occurrence and development of leukemia and is closely related to the treatment and prognosis of leukemia. The standard chemotherapy strategies for the pre-treatment of leukemia have many drawbacks. Hence, the usage of antioxidants and oxidants in the treatment of leukemia is being explored and has been preliminarily applied. This article reviews the research progress of oxidative stress and leukemia. In addition, the application of antioxidants treatment in leukemia has been summarized.

  • 简介:Athree-dimensionalgeometricmodeloftheoxidativecouplingofmethane(OCM)packed-bedreactorloadedwithNa2WO4-Mn/SiO2particulatecatalystwassetup,andanimprovedStanschkineticmodelwasestablishedtocalculatetheOCMreactionsusingthecomputationalfluiddynamicsmethodandFluentsoftware.Thesimulationconditionswerecompletelythesamewiththeexperimentalconditionsthatthevolumevelocityofthereactantwas80mL/minunderstandardstate,theratioofCH4/O2was3,thetemperatureandpressurewere800°Cand1atm,respectively.Thecontourofthecharacteristicsparametersinthecatalystbedwasanalyzed,suchasthespeciesmassfractions,temperature,theheatfluxonsidewallsurface,pressure,fluiddensityandvelocity.TheresultsshowedthatthecalculatedvaluesmatchedwellwiththeexperimentalvaluesontheconversionofCH4andtheselectivitytoproducts(C2H6,C2H4,CO2,CO)inthereactoroutletwithanerrorrangeof±2%.ThemassfractionsofCH4andO2decreasedfrom0.6and0.4inthecatalystbedinletto0.436and0.142intheoutlet,wherethemassfractionsofC2H6,C2H4,COandCO2were0.035,0.061,0.032and0.106,respectively.Duetotheexistenceoflaminarboundarylayer,thecontoursofeachcomponentbentupwardsinthevicinityoftheboundarylayer.ThisOCMreactionwasvolumeincreasereactionandthetotalmolesofproductsweregreaterthanthoseofreactants.Theflowfieldinthecatalystbedmaintainedconstanttemperatureandpressure.Thefluiddensitydecreasedgraduallyfrom2.28kg/m3intheinletofthecatalystbedto2.22kg/m3intheoutletofthecatalystbed,whilethevelocityincreasedfrom0.108m/sto0.115m/s.

  • 简介:AbstractRespiratory health in the general population declines regardless of the presence of pulmonary diseases. Oxidative stress has been implicated as one of the mechanisms involved in respiratory dysfunction. This review was to evaluate studies that relate oxidative stress factors with pulmonary function among the general population without prior respiratory illnesses. The search yielded 54 citations. Twenty-one studies qualified for incorporation in this review. Owing to the heterogeneity of the review, studies were discussed based on identified oxidative stress factors responsible for pulmonary dysfunction. Oxidative stress biomarkers, including gene polymorphisms of nuclear factor erythroid 2-related factor 2, heme oxygenase 1, glutathione S transferase, superoxide dismutase, and lipid peroxidation products were involved in lung function decline. In addition, the antioxidant status of individuals in reference to dietary antioxidant intake and exposure to environmental pollutants affected oxidative stress and pulmonary function, as indicated by forced expired volume in one second, forced vital capacity, and forced expiratory flow at 25%-75%. This review indicated that oxidative stress is implicated in the gradual decline of lung function among the general population, and gene polymorphism along the antioxidant defense line and/or their interaction with air pollutants reduce lung function. Different polymorphic forms among individuals explain why the rate of lung function decline differs among people. Dietary antioxidants have respiratory health benefits in antioxidant gene polymorphic forms. Therefore, the genetic composition of an individual may be considered for monitoring and identifying people at risk of respiratory illnesses.

  • 简介:Oxidativedesulfurization(ODS)ofAzericrudeoilwasstudiedundertheelectricfield.Bymeansoforthogonalexperiments,theoptimumconditionsfordesulfurizationofcrudeoilwereidentified,whiletheeffectofoxidativedesulfurizationundertheelectricfieldwascomparedwiththatofthesingleoxidativedesulfurizationmethod.Thetestresultsshowedthatthedesulfurizationreactiontakingplaceundertheelectricfieldwouldbebetterthanthetraditionalmethod.Thedesulfurizationrateofcrudeoilcouldreachupto77.06%at115℃atadistilledwateramountof10m%,adesulfurizerdosageof200μg/g,andademulsifierdosageof50μg/g.

  • 简介:AIM:NitrativeandoxidativeDNAdamagesuchas8-nitroguanineand8-oxo-7,8-dihydro-2'-deoxyguanosine(8-oxodG)formationhasbeenimplicatedininitiationand/orpromotionofinflammation-mediatedcarcinogenesis.TheaimofthisstudyistoclarifywhethertheseDNAlesionsparticipateintheprogressionofintrahepaticcholangiocarcinoma.METHODS:Weinvestigatedtherelationoftheformationof8-nitroguanineand8-oxodGandtheexpressionofhypoxia-induciblefactor-1α(HIF-1α)withtumorinvasionin37patientswithintra-hepaticcholangiocarcinoma.RESULTS:Immunohistochemicalanalysesrevealedthat8-nitroguanineand8-oxodGformationoccurredtoamuchgreaterextentincanceroustissuesthaninnon-canceroustissues.HIF-1αcouldbedetectedincanceroustissuesinallpatients,suggestinglowoxygentensioninthetumors.HIF-1αexpressionwascorrelatedwithinduciblenitricoxidesynthase(iNOS)expression(r=0.369andP=0.025)and8-oxodGformation(r=0.398andP=0.015).DoubleimmunofluorescencestudyrevealedthatiNOSandHIF-1αco-localizedincanceroustissues.Notably,theformationof8-oxodGwascorrelatedsignificantlywithlymphaticinvasion(r=0.386andP=0.018).Moreover,8-nitroguanineand8-oxodGinnon-canceroustissueswereassociatedsignificantlywithneuralinvasion(P=0.042andP=0.026,respectively).TheseresultssuggestthatreciprocalactivationbetweenHIF-1αandiNOSmediatespersistentDNAdamage,whichinducestumorinvasivenessviamutations,resultinginpoorprognosis.CONCLUSION:Theformationof8-nitroguanineand8-oxodGplaysanimportantroleinmultiplestepsofgeneticchangesleadingtotumorprogression,includinginvasiveness.

  • 简介:Oxidativestressiscloselyassociatedwithsecondarycelldeathinmanydisordersofthecentralnervoussystemincludingstroke,Parkinson’sdisease,Alzheimer’sdisease.Amongmanyaberrantoxidativestress-associatedproteins,DJ-1hasbeenassociatedwiththeoxidativestresscelldeathcascadeprimarilyinParkinson’sdisease.Althoughprincipallyexpressedinthecytoplasmandnucleus,DJ-1canbesecretedintotheserumunderpathologicalcondition.Recently,aclosepathologicalassociationbetweenDJ-1andoxidativestressinstrokehasbeenimplicated.Tothisend,weandothershavedemonstratedtheimportantroleofmitochondriainneuroprotectionforstrokebydemonstratingthatthetranslocationofDJ-1inthemitochondriacouldpotentiallymitigatemitochondrialinjury.Here,wediscussourrecentfindingstestingthehypothesisthatDJ-1notonlyfunctionsasaformofintracellularprotectionfromoxidativestress,butthatitalsoutilizesparacrineand/orautocrinecuesinordertoaccomplishextracellularsignalingbetweenneighboringneuronalcells,resultinginneuroprotection.ThisarticlehighlightsrecentevidencesupportingthestatusofDJ-1askeyanti-oxidativestresstherapeutictargetforstroke.

  • 简介:AbstractObjective:Clinically, low-dose aspirin and progesterone are frequently used to prevent pregnancy loss. We investigated the effect of these drugs on the biological behavior of human extravillous trophoblasts in vitro.Methods:HTR-8/SVneo cells were cultured in vitro and treated with different concentrations of aspirin and progesterone. The proliferation, invasion, and apoptosis of HTR-8/SVneo cells were assessed using a cell counting Kit-8 assay, Matrigel Transwell assay, and Hoechst staining, respectively. Reverse transcriptase polymerase chain reaction was used to verify the expression of related genes. Reactive oxygen species (ROS) levels were detected using the 2,7-dichlorofluorescin diacetate assay.Results:Low-dose aspirin alone, progesterone alone, or aspirin plus progesterone upregulated the proliferation and invasion and decreased the apoptosis of HTR-8/SVneo cells. Moreover, the expression of marker of proliferation Ki-67 (MKI67), matrix metalloproteinases 2 (MMP2), and MMP9 was increased. In addition, low-dose aspirin plus progesterone exerted stronger anti-apoptosis effects than low-dose aspirin and progesterone alone. Interestingly, aspirin upregulated the expression of progesterone receptor (PGR). Treatment with hydrogen peroxide (H2O2) promoted ROS production in HTR-8/SVneo cells; however, low-dose aspirin plus progesterone significantly restricted H2O2-mediated ROS production and apoptosis in HTR-8/SVneo cells.Conclusions:These data suggest that low-dose aspirin and progesterone promote proliferation and invasion and cooperatively reduce oxidative stress and apoptosis in trophoblasts in vitro. These results may provide an experimental basis for the combined application of aspirin and progesterone to prevent unexplained recurrent spontaneous miscarriage, especially in patients with trophoblast dysfunction.

  • 简介:Zygosaccharomycesrouxiiisasalt-tolerantyeastspeciescapableofremovingcadmium(Cd)pollutantfromaqueoussolution.Presently,thephysiologicalcharacteristicsofZ.rouxiiunderthestressofsodiumchloride(NaCl)andCdarepoorlyunderstood.ThisstudyinvestigatedtheeffectsofNaClandCdonthegrowth,oxidativestressandantioxidantenzymeactivitiesofZ.rouxiiafterstresstreatmentfor24h.ResultsshowedthatNaClorCdalonenegativelyaffectedthegrowthofZ.rouxii,butthegrowth-inhibitingeffectofCdonZ.rouxiiwasreducedinthepresenceofNaCl.FlowcytometryassayshowedthatunderCdstress,NaClsignificantlyreducedtheproductionofreactiveoxygenspecies(ROS)andcelldeathofZ.rouxiicomparedwiththoseintheabsenceofNaCl.Theactivitiesofsuperoxidedismutase(SOD),catalase(CAT),andperoxidase(POD)ofZ.rouxiiweresignificantlyenhancedby2%–6%NaCl,whichlikelycontributedtothehighsalttoleranceofZ.rouxii.ThePODactivitywasinhibitedby20mgL-1CdwhiletheSODandCATactivitieswereenhancedby8mgL-1Cdandinhibitedby20mgL-1or50mgL-1Cd.Theinhibitoryeffectofhigh-levelCdontheantioxidantenzymeactivitiesofZ.rouxiiwascounteractedbythecombineduseofNaCl,especiallyat6%.ThisprobablyaccountedforthedecreaseinCd-inducedROSproductionandcelldeathofZ.rouxiiafterincubationwithNaClandCd.OurworkprovidedphysiologicalcluesastotheuseofZ.rouxiiasabiosorbentforCdremovalfromseawaterandliquidhighlysaltyfood.
