  • 简介:摘要目的探讨血清miRNA-145(miR-145)、miRNA-21(miR-21)和miRNA-197(miR-197)对非小细胞肺癌(NSCLC)的诊断价值。方法回顾性分析2019年1月至2020年6月四川省泸州市中医医院收治的65例NSCLC患者的临床资料,以60名健康体检者为健康对照组。采用实时荧光定量聚合酶链反应(qRT-PCR)检测所有受试者血清miR-145、miR-21和miR-197的相对表达量,分析其与NSCLC患者临床病理特征的关系。采用受试者工作特征(ROC)曲线分析miR-145、miR-21和miR-197单独及联合诊断NSCLC的效能。结果NSCLC组miR-145相对表达量低于健康对照组(1.05±0.40比1.38±0.44,t=-4.326,P<0.01),miR-21、miR-197相对表达量均高于健康对照组(2.37±0.89比0.83±0.25,4.42±0.75比1.10±0.33,均P<0.01)。NSCLC组中,TNM分期Ⅲ~Ⅳ期患者血清miR-21和miR-197相对表达量均高于Ⅰ~Ⅱ期患者(均P<0.01),腺癌患者miR-145相对表达量高于鳞状细胞癌患者(P<0.01),淋巴结转移患者miR-21相对表达量高于无淋巴结转移患者(P<0.01)。miR-145、miR-21、miR-197单独诊断NSCLC时,曲线下面积(AUC)分别为0.842(95% CI 0.795~0.907)、0.868(95% CI 0.812~0.938)、0.857(95% CI 0.801~0.913),诊断最佳截断值分别为1.14、2.03、2.98;采用最佳截断值进行诊断时,单独检测灵敏度最高的是miR-145(83.76%),特异度最高的是miR-21(87.63%);三者联合检测时AUC为0.939(95% CI 0.904~0.965),诊断灵敏度为91.58%,特异度为79.86%。结论miR-145、miR-21、miR-197相对表达量与NSCLC患者病理类型、TNM分期、淋巴结转移相关,可能成为辅助诊断NSCLC的标志物,三者联合检测可提高诊断效能和灵敏性。

  • 简介:摘要目的探讨外周血miRNA-146a、miRNA-155和miRNA-186在重症肺炎患者中的表达及临床意义。方法选择宁波市医疗中心李惠利医院东部院区2019年2月至2021年2月收治的重症肺炎患者79例为研究对象,按照入院28 d预后情况,分为存活组(58例)与死亡组(21例);另选择同时段健康体检者60例为对照组。采集研究对象外周血标本,分离血清,采用实时荧光定量PCR法(qRT-PCR)测定外周血miRNA-146a、miRNA-155和miRNA-186表达。结果重症肺炎组急性生理学和慢性健康状态Ⅱ(APACHE Ⅱ)评分[(18.54±2.83)分]高于对照组[(3.18±0.57)分](t=41.37,P < 0.05)。重症肺炎组外周血miRNA-146a相对表达量(0.32±0.07)低于对照组(1.08±0.21),而miRNA-155相对表达量(2.54±0.46)和miRNA-186相对表达量(3.16±0.38)均高于对照组[(0.42±0.09)、(0.89±0.17)](t=30.07、35.16、43.08,均P < 0.001)。死亡组外周血miRNA-146a相对表达量(0.25±0.68)低于存活组(0.59±0.12),而miRNA-155相对表达量(3.97±0.78)和miRNA-186相对表达量(5.23±0.86)均高于存活组[(0.89±0.21)、(1.52±0.23)](t=19.11、27.69、30.29,均P < 0.001)。APACHE Ⅱ评分与miRNA-146a呈线性负相关(r=-0.75,P=0.015),而与miRNA-155和miRNA-186呈线性正相关(r=0.82、0.70,P=0.002、0.021)。重症肺炎诊断中,miRNA-146a灵敏度为72.34%,特异度为62.50%;miRNA-155灵敏度为75.00%,特异度为62.96%;miRNA-186灵敏度为66.67%,特异度为48.65%。结论外周血miRNA-146a在重症肺炎中低表达,而miRNA-155和miRNA-186在重症肺炎中高表达,且miRNA-146a、miRNA-155和miRNA-186与患者预后密切相关。

  • 简介:MicroRNA(miRNA)是一类广泛存在于人、动植物和病毒的内源长度约为21~25个核苷酸的非编码小RNA,且在不同组织、不同阶段存在特异性表达,在真核生物发育和基因表达中通过与靶mRNA形成完全或不完全互补配对从而扮演着重要角色。参与动物体发育、细胞增殖与死亡、细胞分化等各种过程。文章综述了miRNA对家禽发育生长、脂质代谢、疾病控制等方面的研究进展,为进一步研究miRNA在家禽中功能提供理论依据和事实参考。

  • 简介:摘要:miRNA在基因调控方面起着重要作用,其异常表达和特异性表达提示其可成为诊断或预后的生物标志物。因此准确测定其表达水平对于其应用十分关键。但是由于miRNA序列短,同源序列相似度高,含量低,缺乏能够使其选择性扩增的共同特征,使得检测miRNA十分具有挑战性。本文对近年来miRNA的检测方法进行了概述,包括传统方法及其改进进展,以及一些新的检测方法,并对这些方法的优缺点进行了总结。

  • 简介:SmallinterferingRNA(siRNA)andmicroRNA(miRNA)aresmallRNAsof18-25nucleotides(nt)inlengththatplayimportantrolesinregulatinggeneexpression.TheyareincorporatedintoanRNA-inducedsilencingcomplex(RISC)andserveasguidesforsilencingtheircorrespondingtargetmRNAsbasedoncomplementarybase-pairing.ThepromiseofgenesilencinghasledmanyresearcherstoconsidersiRNAasananti-viraltool.However,inlong-termsettings,manyvirusesappeartoescapefromthistherapeuticalstrategy.Anexampleofthismaybeseeninthecaseofhumanimmunodeficiencyvirustype-1(HIV-1)whichisabletoevadeRNAsilencingbyeithermutatingthesiRNAtargetedsequenceorbyencodingforapartialsuppressorofRNAi(RNAinterference).Ontheotherhand,becausemiRNAtargetingdoesnotrequireabsolutecomplementarityofbase-pairing,mutationalescapebyvirusesfrommiRNAspecifiedsilencingmaybemoredifficulttoachieve.Inthisreview,wediscussstratagemsusedbyvariousvirusestoavoidthecells'antiviralsi/mi-RNAdefensesandnotionsofhowvirusesmightcontrolandregulatehostcellgenesbyencodingviralmiRNAs(vmiRNAs).
