摘要
Linkerswereassembledonaglasssurfacebasedonthehydrolysisandcondensationof3-glycidoxy-propyltrimethoxysilane(GPS).AftertheassemblyofGPS,fourapproachesweretriedtoopentheendingepoxidegroupofGPSortofurtherelongatethelinkers.TheeffectoftheseapproachesonDNAinsitusynthesisandhybridizationwasinvestigated.Forthespacingofthesynthesisinitiationsites,thewettabilityofthesupportandthelengthofthelinkinggroupthatattachestheinitiationsitetothesurfacehavedirectinfluencesontheyieldofcouplingreactionsandthesubsequenthybridizationevents.X-rayphotoelectronspectroscopy(XPS)andmeancontactanglesofdeionizedwateroftheaboveslidesweremeasuredtoassessthelinker'scharacteristicsineachprocedure.Itwasprovedthattheglassslidesweresuccessfullymodifiedandbecameexcellentsupportsfortheoligonucleotidessynthesis.Inaddition,itprovedbestfortheinsituoligonueleotidessynthesisthataglassslidewasinturntreatedwithethylenediamine,glutaradehyde,ethanolamineandsodiumborohydridesolutionatambienttemperatureaftersilanizedwithGPS.
