Inthisstudy,28,691genomesequencesand16,566expressedsequencetags(ESTs)ofEucalyptuswerederivedfromtheGenBankdatabase.Atotalof2292SSRlociweresoughtoutfrom1785effectivesequences.ThroughanalysesofSSRlociinformation,theSSRmotiflengthwasnegativelycorrelatedwiththeabundanceoftheSSRs.IntheESTsequencesofEucalyptus,tripletrepeatmotifswerethemostabundant,anddinucleotiderepeatsmotifshadthehighestfrequencies.Subsequently,395pairsofprimersweredesignedbasedontheSSRloci.UsingoptimizedSSR-PCRconditions,340pairsofprimersweresuccessfullyscreened,withasuccessrateof86.1%.Byconstructionofamaximumlikelihoodphylogenetictreeofsixeucalyptspecies,representedbyfivespeciesofthegenusEucalyptusandoneofthegenusCorymbia,thegeneticrelationshipsofEucalyptusurophyllaandE.camaldulensissuggestedbythistreewasfoundtodifferfromthatsuggestedbytraditionalmorphologicaltaxonomy.TheresultsprovideinsightsforevaluatinggeneticpersityofEucalyptusandanalysisofEucalyptusphylogeneticsusingSSRmarkers.
