  • 简介:Cochlearouterhaircells(OHCs)areinvolvedinamechanicalfeedbackloopinwhichthefastsomaticmotilityofOHCsisrequiredforcochlearamplification.Alternatively,amplificationisthoughttoarisefromactivehairbundlemovementsobservedinnon-mammalianhaircells.Wemeasuredthevoltage-evokedhairbundlemotionsinthegerbilcochleatodetermineifsuchmovementsarealsopresentinmammalianOHCs.TheOHCsdisplayedalargehairbundlemovementthatwasnotbasedonmechanotransducerchannelsbutbasedonsomaticmotility.Significantly,bundlemovementswereabletogenerateradialmotionofthetectorialmembraneinsitu.Thisresultimpliesthatthemotility-associatedhairbundlemotionmaybepartofthecochlearamplifier.

  • 简介:Phaeodactylumtricornutumisoneoftheimportantmarinediatomsforoceanicprimaryproduction.Itsreproductionhasprofoundsignificanceinthelifecycle;however,thenuclearbehaviorduringitssexualreproductionwasnotclear.Inthisstudy,weobservedthenucleartransitionanddetermineditscorrelationwithcellconjunction.Itwasfoundthattwocellsjointedattheirapicesfirstandswungandalignedeachotherimmediately,andnucleusfromonecellwasabletotransferintoanotheroneduringcellconjugation.Thecellpairsconjugatedfornucleartransitionweredifferentfromthoseformedinmitosisinhypovalvethicknessandcellulararrangement.OurfindingsprovedtheexistenceofsexualreproductioninP.tricornutum.

  • 简介:Objective:ToconfirmwhetherBungarusmulticinctuscrudevenominducestheapoptosisofK562tumorcellsandtofindoutthecomponentsinducingapoptosisofK562cellsfromthecrudevenom.Methods:thecrudevenomseparatedandpurifiedbycationexchangechromatography,andtheeffectofvenomsonK562wasstudiedbyMTTmethodandflowcytometry.Results:ThecrudevenombegantokillK562cellsatthan8(103ng/ml(thesurvivalratewas82.5%)concentrationandtheeffectwasmoresignificantin24hwhenadministrating8(105ng/ml(thesurvivalratewas29.4%)crudevenom.ApoptoticbodieswereobservedintheK562tumorcellsbyfluorescentmicroscopyafteradministrationof5(g/mlcycloheximide(CHX)orthepeakVIsolutionatabout8(105ng/ml.Thesameresultsweredetectedbytheflowcytometry.Asub-G1peakappearedafteradministrationofCHXorthesixthpeaksolution.Conclusion:TheauthorsfoundthatthevenomcankillK562tumorcellsintime-anddose-dependentmanner.However,thekillingeffectofthevenomisnotapoptosis.What'smore,thepeakVIsolution,acomponentofthecrudevenomcaninducetheapoptosisofK562tumorcells.

  • 简介:AbstractBackground:Circular RNA (circRNA) is a type of closed circular noncoding RNA (ncRNA), mostly formed by back-splicing or alternative splicing of pre-messenger RNA (mRNA). The aim of this study was to explore the expression profile of circRNA in peripheral blood mononuclear cells (PBMCs) of patients with ankylosing spondylitis (AS) and discover potential molecular markers of AS.Methods:The circRNA microarray technology was used to detect the expression of circRNAs in the peripheral blood of 6 patients with AS and 6 healthy controls (HC). To screen the differentially expressed circRNAs by fold change (FC) and P value, these differentially expressed circRNAs were analyzed by bioinformatics. In 60 cases of AS and 30 cases of HC, 4 circRNAs were subjected to real-time fluorescence quantitative polymerase chain reaction (RT-qPCR), and their correlation with various clinical indicators was analyzed. Finally, the receiver operating characteristic (ROC) curve was used to analyze their potential as AS diagnostic markers.Results:The microarray results showed that there were 1369 significantly differently expressed (P < 0.05, FC > 1.5) circRNAs between the AS and HC groups (675 upregulated and 694 downregulated). The results of bioinformatics analysis suggested that they were mainly involved in "enzyme binding," "adenosine ribonucleotide binding," "MAPK signaling pathway" , etc. The RT-qPCR results showed that the expressions of hsa_circRNA_001544 (U = 486.5, P < 0.05) and hsa_circRNA_102532 (U = 645, P < 0.05) were significantly different between the AS group and the HC group. The AS group was further divided into two subgroups: active AS (ASA) and stable AS (ASS). After analysis, it was found that compared with the HC group, hsa_circRNA_001544 was significantly increased in both ASA (U = 214, P < 0.05) and ASS groups (U = 273, P < 0.05), while hsa_circRNA_008961 (U = 250, P < 0.05) and hsa_circRNA_102532 (U = 295, P < 0.05) were only significantly increased in the ASA group. Furthermore, hsa_circRNA_012732 was significantly different between the ASA and ASS groups (U = 194, P < 0.05), and there was no statistical significance among the remaining groups. Correlation analysis results showed that hsa_circRNA_012732 was negatively correlated with Bath Ankylosing Spondylitis Disease Activity Index (BASDAI), high-sensitivity C-reactive protein (hsCRP), and globulin (GLOB) and positively correlated with lymphocyte count (LY), mean corpusular volume, and albumin (ALB), and hsa_circRNA_008961 was negatively correlated with platelet (PLT) count. ROC curve analysis showed that hsa_circRNA_001544 (95% CI = 0.610-0.831, P < 0.05) and hsa_circRNA_102532 (95% CI = 0.521-0.762, P < 0.05) were statistically significant, and their area under curve (AUC) values were 0.720 and 0.642, respectively.Conclusions:There are differentially expressed circRNAs in PBMCs of AS patients, and they may be involved in the occurrence and development of AS. Among these differentially expressed circRNAs, hsa_circRNA_012732 has the potential to become an indicator of disease activity, and hsa_circRNA_001544 has the potential to become a molecular marker for AS diagnosis.

  • 简介:AbstractThe effects of gestational diabetes mellitus (GDM) on offspring include macrosomia, hypoglycemia, respiratory distress syndrome, cardiovascular disease, neural and mental injury, etc. The effects of GDM on the health status of offspring are sustained although pregnancy has ended. It has been proposed that fetal reprogramming causes long-term consequences to metabolic health in offspring. An intrauterine high-glucose environment may lead to changes in the multi-differentiation proficiency of intracorporal stem cells, showing decreased proliferation and osteogenic ability, increased adipogenic ability, accelerated apoptosis, and occurrence of premature failure. This environment also reduces the mobilization of bone marrow stem cells, whereas it increases that of medullary cells. This results in pro-inflammatory conditions and sustained inflammation in the body, thereby increasing the risk of obesity, cardiovascular and neurological disorders, and metabolic abnormalities. Stem cells derived from the amniotic membrane, umbilical cord, or placenta may be a reliable predictor of the long-term effects of GDM on offspring. The levels of blood glucose during pregnancy should be effectively controlled to reduce harm to the neonate.

  • 简介:Objective:Toobservetheeffectsofthermalstressonproliferationofhumanvascularendothelialcells(VECs)andexploreitssignificance.Methods:ChangesofVECsproliferationwereinvestigatedwith3H-TdRincorporationmethodafterECV304wastreatedat43℃for2hours,whileexpressionsofintercellularadhesionmolecule-1(ICAM-1),inhibitorofdifferentiation-1(ID1),andP16andP21proteinsweredeterminedbyWesternBlotting.Results:TheeffectofinhibitionofVECsgrowthafterthermalstresswasdetectedby3H-TdRincorporationexperiment.WesternblottingshowedICAM-1,amarkerofactivatedendothelialcells,wasincreasedmarkedlyafterthermalstress.ExpressionofID1proteindeclinedgraduallywithincreasingexpressionsofitsdownstreamgenes,P16andP21followingthethermalstress.Conclusions:ThermalstresscouldstronglyactivateVECsandinhibitproliferationofVECsthroughID1,thusdownregulatingcyclin-dependentkinaseinhibitors,P16andP21,whichmightbeanessentialpathwayforrecoveryofVECsafterthermalstress.

  • 简介:Swelling-activatedCl-currents,I(Cl,swell),weremeasuredduringhyposmoticshockinwhiteLeghornembryonicchickheartcellsusingthewhole-cellrecordingofpatch-clamptechnique.Genistein,aninhibitorofproteintyrosinekinase(PTK),suppressedI(Cl,swell).Underisosmoticconditionphorbol12-myristate13-acetate(PMA),anactivatorofPKC,elicitedtheCl-currentsimilartothatinhyposmoticsolution,whereashyposmoticshockdidnotelicitI(Cl,swell)inchelerythrinechloride(aninhibitorofPKC)-treatedcells.Con-focalmicroscopyexperimentsusingFITC-phalloidinasafluorescentlabelofF-actinshowedthattheactinnetworkwasmovedfromcorticalregionofthecelltothecenterafterhyposmoticshockascomparedwiththeimageunderisosmoticcondition.WhenthecellsweretreatedwithcytochalasinB(CB)orcytochalasinD(CD)underisosmoticconditionthedisruptionoftheF-actinintegritywasobserved,andI(Cl,swell)wasnotelicited.WithcombinationtreatmentofCBwithPMA,hyposmoticsolutioncouldnotelicitedI(Cl,swell).TheresultssuggestedthattheroleofPTK,probablyreceptortyrosinekinase,forregulationofI(Cl,swell)appearedtobeatupstreamsiterelatedtotheroleofF-actin.ThenPKCsignalpathwaywasactivatedsomehowandfinallychangeinthepolymerizationstateofcytoskeletonledtoactivatetheswelling-activatedCl-channels.TheseresultsdemonstrateclearlythatPTK,PKCandF-actinareimportantfactorsforreg-ulationofI(Cl,swell),inembryonicchickheartcellsascomparedwithoftencontroversialresultsreportedindifferentcelltypes.

  • 简介:AbstractInnate lymphoid cells (ILCs) are a group of lymphocytes without diversified antigen receptors encoded by gene rearrangement on T and B cells. ILCs, which are tissue-resident innate immune cells, expressed particularly in the mucosa or the barrier surface, contribute to the formation of lymphoid organs, the maintenance of tissue homeostasis, and the regulation of antimicrobial defenses. It has been recently reported that ILCs were enriched at the maternal-fetal interface. During a successful pregnancy, the maternal immune system must tolerate a fetus as an allograft. With the new defined of ILCs, a number of studies have shown that three types of ILCs are involved in embryonic development and pregnancy maintenance as well as the occurrence and development of pregnancy-related complications. This article reviews the types and roles of ILCs in normal pregnancy and pregnancy-related diseases.

  • 简介:TheeffortonelectrochemicalreductionofCO2tousefulchemicalsusingtherenewableenergytodrivetheprocessisgrowingfastrecently.Inthisreview,weintroducetherecentprogressesontheelectrochemicalreductionofCO2insolidoxideelectrolysiscells(SOECs).Athightemperature,onlyCOisproducedwithhighcurrentdensitiesandFaradicefficiencywhilethereactoriscomplicatedandabettersealingtechniqueisurgentlyneeded.Thetypicalelectrolytessuchaszirconia-basedoxides,ceria-basedoxidesandlanthanumgallates-basedoxides,anodesandcathodesareintroducedinthisreview,andthecathodematerials,suchasconventionalmetal–ceramics(cermets),mixedionicandelectronicconductors(MIECs)arediscussedindetail.Inthefuture,togainmorevalue-addedproducts,theelectrolyte,cathodeandanodematerialsshouldbedevelopedtoallowSOECstobeoperatedattemperaturerangeof573–873K.Atthosetemperatures,SOECsmaycombinetheadvantagesofthelowtemperaturesystemandthehightemperaturesystemtoproducevariousproductswithhighcurrentdensities.
