  • 简介:Objective:Todeterminetheefficacyofpolylacticacidglueinpreventingepiduralscaradhesionafterlaminectomyinrabbits.Methods:Twenty-fourJapanesewhiterabbitsunderwentlaminectomy(includingtheattachedligaments)atL2andL5.AfterlaminectomyatL5,polylacticacidgluewassprayedontheduraandnerverootsandthissegmentwastakenastheexperimentalgroup.AfterlaminectomyatL2,nothingwasusedandthissegmentwasenrolledastheselfcontrolgroup.Fourrabbitswerekilledeverytwoweekspostoperativelytilltheendoftheexperimentat12weeks.Thentheoperatedspinewasobservedgrossly,histologicallyandultrastructurallytocheckthedegreeofscarformation,thestatusofepiduralscaradhesion,theabsorptionoftheglue,andtheintracellularstructureoffibroblasts.Results:Thegluecoagulatedimmediatelyaftersprayingandshowedexcellenthemostaticeffect.Thegluemembranewaseasytobetakenawayfromtheduramaterofthesamplesfor2weeksandtherewerenocellsintheepiduralspaceintheexperimentalgroup.Buttheduramaterwascoveredbyhematomainthecontrolgroup,whichformedmildadhesion,withfibroblastsproliferatingactively.Inthe4thweek,someglueshiversremainedintheepiduralspacewithfibroblastsincreasingalittle,andtheduramaterwassmoothintheexperimentalgroup.However,inthecontrolgroup,theformedscarwasfragileandconglutinatedwiththeduramaterdiffuselyandfibroblastsweremuchmorethanthoseintheexperimentalgroup.Inthe6th-12thweeks,therewasapotentialinterspacebetweenthescarandtheduramater,andthepolylacticacidgluewasabsorbedcompletelyintheexperimentalgroup.Muchtoughscarwasfoundinthecontrolgroup,whichwasverydifficulttodissectfromtheduramaterandthesurroundingtissues.Fromtheultrastructuralobservationofthefibroblasts,thenucleusbecamemuchbiggerandtheroughendoplasmicreticulumwasmuchmoreplentifulinthecontrolgroupthanthatinthee

  • 简介:Objective:ToexplorethepreparativemethodandstudythedegradationcharacteristicsofbonerepaircompositeofDL-polylacticacid(PDLLA)/hydroxyapatite(HA)/decalcifyingbonematrix(DBM)invitro.Methods:AnemulsionblendmethodwasdevelopedtopreparethecompositeofPDLLA/HA/DBMinweightratioofPDLLA:HA:DBM=1.5-2:1-1.5:1.Thedynamicchangesofweight,biomechanicalpropertyandpHvalueofPDLLA/HA/DBMandPDLLAinphosphatebufferedsaline(PBS,pH7.4)werestudiedrespectivelythroughdegradationtestsinvitro.Results:Withoutbeingheated,PDLLA,HAandDBMcouldbesynthesizedwiththeemulsionblendmethodasbonecompositeofPDLLA/HA/DBM,whichhadbothosteoconductiveandosteoinductiveeffects.Thediameteroftheaperturewas100-400μmandthegapratewas71.3%.Duringdegradation,thepHvalueofPDLLAsolutiondecreasedlightlywithin2weeks,butdecreasedobviouslyattheendof4weeksandthevaluewas4.0.WhilethepHvalueofPDLLA/HA/DBMkeptquitesteadyandwas6.4attheendof12weeks.TheweightofPDLLAchangedlittlewithin4weeks,thenchangedobviouslyandwas50%ofitsinitialweightattheendof12weeks.WhiletheweightofPDLLA/HA/DBMchangedlittlewithin5weeks,thenchangedobviouslyandwas60%oftheinitialweightattheendof12weeks.TheinitialbiomechanicalstrengthofPDLLAwas1.33MPa,decreasedlittlewithin3weeks,thenchangedobviouslyandkeptat0.11MPaattheendof12weeks.TheinitialbiomechanicalstrengthofPDLLA/HA/DBMwas1.7MPa,decreasedlittlewithin4weeks,thenchangedobviouslyandkeptat0.21MPaattheendof12weeks.Conclusions:Theemulsionblendmethodisanewmethodtopreparebonerepairmaterials.Asanewbonerepairmaterial,PDLLA/HA/DBMismoresuitableforregenerationandcellimplantation,andtheenvironmentduringitsdegradationisadvantageoustothegrowthofbonecells.

  • 简介:Objective:Impairedactivefluidtransportofalveolarepitheliummayinvolveinthepathogenesisandresolutionofalveolaredema.Thcobjectiveofthisstudywastoexplorethechangesinalveolarepithelialliquidclearanceduringlungedemafollowingacutelunginjuryinducedbyoleicacid.Methods:Forty-eightWistarratswererandomlydividedintosixgroups,I.e.,injured,amiloride,ouabain,amilorideplusouabainandterbutalinegroups.Twenty-fourhoursaftertheinductionofacutelunginjurybyintravenousoleicacid(0.25ml/kg),5%albuminsolutionwith1.5μCi125Ⅰ-labeledalbumin(5ml/kg)wasdeliveredintobothlungsviatrachea.Alveolarliquidclearance(ALC),extravascularlungwater(EVLW)contentandarterialbloodgasesweremeasuredonehourthereafter.Results:At24haftertheinfusionofoleicacid,theratsdevelopedpulmonaryedemaandseverehypoxemia,withEVLWincreasedby47.9%andALCdecreasedby49.2%.Additionofeither2×10-3Mamilorideor5×10-4MouabaintotheinstillationfurtherreducedALCandincreasedEVLW.ALCincreasedbyapproximately63.7%andEVLWdecreasedby46.9%withimprovedhypoxemiaintheTerbutaline(10-4M)group,comparedthoseininjuredrats.AsignificantnegativecorrelationwasfoundbetweentheincrementofEVLWandthereductionofALC.Onclusions:Activefluidtransportofalveolarepitheliummightplayaroleinthepathogenesisoflungedemainacutelunginjury.
