  • 简介:ThepresentstudywasdesignedtodeterminetheeffectsofatraditionalChinesemedicine,calledQishenYiqiDroppingPillonchronichypoxia-inducedmyocardialinjury.ToestablisharatchronichypoxiamodeltobeusedintheevaluationofthetherapeuticeffectsoftheQishenYiqiDroppingPill,Sprague-Dawley(SD)ratswererandomlydividedintothreegroups:thecontrol,model,andtreatmentgroups(n=10pergroup).Theanimalswerehousedinaplexiglasscontainer.Thecontrolanimalswereundernormaloxygenconcentrationandthemodelandtreatmentgroupswereexposedtoairandnitrogenfor5weeks.TheratsinthetreatmentgroupwereorallyadministeredtheQishenYiqiDroppingpill(35mg·kg-1·d-1)for5weeks.Afterthetreatment,thecardiacfunctionandmorphologywereanalyzed,andtheexpressionlevelsofhypoxia-induciblefactor1α(HIF-1α)weredeterminedusingWesternblotting.Ourresultsindicatedthatthecardiacfunctionwasimpaired,cellapoptosiswasenhanced,andHIF-1αexpressionwasup-regulatedinthemodelgroup,comparedtothecontrolgroup.ThesechangeswereamelioratedbythetreatmentwiththeQishenYiqiDroppingPill.Inconclusion,QishenYiqiDroppingpillcanamelioratemyocardialinjuryinducedbychronichypoxia,improvecardiacfunction,anddecreasemyocardialcellapoptosis,whichmayprovideabasisforitsclinicaluseforthetreatmentofchroniccardiovasculardiseases

  • 简介:Sheng-Mai-San(SMS),awell-knownChinesemedicinalplantformula,iswidelyusedforthetreatmentofcardiacdiseasescharacterizedbydeficiencyofQiandYinsyndrome.Amousechronicintermittenthypoxia(CIH)modelwasestablishedtomimictheprimaryclinicalfeaturesofdeficiencyofQiandYinsyndrome.MiceexperiencedCIHfor28days(nadir7%topeak8%oxygen,20minperday),resultinginleftventricle(LV)dysfunctionandstructureabnormalities.AfteradministrationofSMS(0.55,1.1,and5.5g·kg-1·d-1)forfourweeks,improvedcardiacfunctionwasobserved,asindicatedbytheincreaseintheejectionfractionfromtheLVonechocardiography.SMSalsopreservedthestructuralintegrityoftheLVagainsteccentrichypotrophy,tissuevacuolization,andmitochondrialinjuryasmeasuredbyhistology,electronmicroscopy,andultrasoundassessments.Mechanistically,theantioxidanteffectsofSMSweredemonstrated;SMSwasabletosuppressmitochondrialapoptosisasindicatedbythereductionofseveralpro-apoptoticfactors(Bax,cytochromec,andcleavedcaspase-3)andup-regulationoftheanti-apoptosisfactorBcl-2.Inconclusion,theseresultsdemonstratethatSMStreatmentcanprotectthestructureandfunctionoftheLVandthattheprotectiveeffectsofthisformulaareassociatedwiththeregulationofthemitochondrialapoptosispathway.

  • 简介:Benignprostatichyperplasia(BPH)isanage-relateddiseaseofunknownetiology,characterizedbyprostaticenlargementcoincidentwithdistinctalterationsintissuehistology.Inthepresentstudy,weinvestigatedwhethertriptolidecanpreventtestosterone-inducedprostatichyperplasiainrats.Castrationwasperformedviathescrotalrouteafterurethaneaesthesia.BPHwasinducedinexperimentalgroupsbydailysubcutaneousinjectionsoftestosteronepropionate(TP)fortwoweeks.Triptolidewasadministereddailybyoralgavageatadoseof100and50μg×kg~(-1)for2weeks,alongwiththeTPinjections.Onday14,theanimalswerehumanelykilledbycervicaldislocationafteraesthesia.Prostateswereexcised,weighed,andusedforhistologicalstudies.Testosteroneanddihydrotestosterone(DHT)levelsinserumandprostateweremeasured.Theresultsshowedthattriptolidesignificantlyreducedtheprostateweight,andthetestosteroneandDHTlevelsinboththeserumandprostate.HistopathologicalexaminationalsoshowedthattriptolidetreatmentsuppressedTP-inducedprostatichyperplasia.Inconclusion,triptolideeffectivelyinhibitsthedevelopmentofBPHinducedbytestosteroneinaratmodel.

  • 简介:Ginkgolicacids(GAs),primarilyfoundintheleaves,nuts,andtestaofginkgobiloba,havebeenidentifiedwithsuspectedallergenic,genotoxicandcytotoxicproperties.However,littleinformationisavailableaboutGAstoxicityinkidneysandtheunderlyingmechanismhasnotbeenthoroughlyelucidatedsofar.InsteadofGAsextract,therenalcytotoxicityofGA(15:1),whichwasisolatedfromthetestaofGinkgobiloba,wasassessedinvitrobyusingMDCKcells.TheactionofGA(15:1)oncellviabilitywasevaluatedbytheMTTandneutralreduptakeassays.Comparedwiththecontrol,thecytotoxicityofGA(15:1)onMDCKcellsdisplayedatime-anddose-dependentmanner,suggestingthecellsmitochondriaandlysosomesweredamaged.ItwasconfirmedthatGA(15:1)resultedinthelossofcellsmitochondrialtrans-membranepotential(ΔΨm).Inpropidiumiodide(PI)staininganalysis,GA(15:1)inducedcellcyclearrestattheG0/G1andG2/Mphases,influencingontheDNAsynthesisandcellmitosis.CharacteristicsofnecroticcelldeathwereobservedinMDCKcellsattheexperimentalconditions,asaresultofDNAagarosegelelectrophoresisandmorphologicalobservationofMDCKcells.Inconclusion,thesefindingsmightprovideusefulinformationforabetterunderstandingoftheGA(15:1)inducedrenaltoxicity.

  • 简介:ScutellariaeRadix(SR),therootofScutellariabaicalensisGeorgi,isusedasanantipyreticdrugandhasbeendemonstratedtohaveanti-inflammatoryactivity.SRisdividedintotwospecifications,'KuQin'(KQ)and'ZiQin'(ZQ),foruseagainstdifferentsymptoms(upperenergizerheatorlowerportionofthetripleenergizer),accordingtothetheoryoftraditionalChinesemedicine(TCM).However,differencesintheefficaciesofthesetwospecificationshavenotbeendetermined.Inthepresentstudy,weaimedtocharacterizethedifferencesintheanti-inflammatoryactivitiesbetweenKQandZQandtoexplorehowtheirdifferencesaremanifestedinlipopolysaccharide(LPS)-inducedmacrophages.Ourresultsshowedthat,inRAW264.7cells(amousemacrophagecelllinederivedfromascites),KQandZQdisplayedanti-inflammatoryeffectsbyinhibitingthereleaseofnitricoxide(NO),inducibleNOS(iNOS),andnuclearfactor-κB(NF-κB)inadose-dependentmannerwithoutdistinction.InNR8383cells(aratalveolarmacrophagecellline),KQandZQdisplayedsimilareffectsonNO,iNOS,andNF-κBasseeninRAW264.7cells,butKQshowedahigherinhibitionrateforNOandiNOSthanthatshownbyZQatthesameconcentration.TheseresultsindicatedthatthereweredifferencesinefficacybetweenKQandZQintreatinglunginflammation.OurfindingsprovidedanexperimentalevidencesupportingthedifferentusesofKQandZQinclinic,asnotedinancientherbalrecords.

  • 简介:Theuterinetetaniccontractionanduterinearterybloodflowreductionarepossiblereasonsforprimarydysmenorrhea(PD).Inthepresentstudy,weaimedtoevaluatetheuterinerelaxanteffectandtheinfluenceonuterinearterybloodvelocityofGe-GenDecoction(GGD),awell-knownChineseherbalformula.InfemaleICRmice,uterinecontractionwasinducedbyoxytocinexposurefollowingestradiolbenzoatepretreatment,andtheuterinearterybloodvelocitywasdetectedbyDopplerultrasound.HistopathologicalexaminationoftheuterinetissuesampleswereperformedbyH&Estaining.Exvivostudiesdemonstratedthatoxytocin,posteriorpituitary,oracetylcholineinducedcontractionsinisolatedmouseuterus.GGDinhibitedbothspontaneousandstimulatedcontractions.InvivostudydemonstratedthatGGDsignificantlyreducedoxytocin-inducedwrithingresponseswithamaximalinhibitionof87%.FurtherstudydemonstratedthatGGDnormalizedoxytocin-inducedabnormalitiesofprostaglandinsF2alpha(PGF2α)andCa2+inmice.Inaddition,injectionofoxytocininducedadecreaseinuterinearterybloodflowvelocity.PretreatmentwithGGDreversedtheoxytocinresponseonbloodflowvelocity.HistopathologicalexaminationshowedpretreatmentwithGGDalleviatedinflammationandedemaintheuteruswhencomparedwiththemodelgroup.BothexvivoandinvivoresultsindicatedthatGGDpossessedasignificantspasmolyticeffectonuterinetetaniccontractionaswellasimprovementonuterinearterybloodvelocitywhichmayinvolvePGF2αandCa2+signaling,suggestingthatGGDmayhaveaclinicpotentialinPDtherapy.
