  • 简介:Cisplatin,awidelyusedanticancerdrug,damageshaircellsincochlearorganotypicculturesatlowdoses,butparadoxicallycauseslittledamageathighdosesresultinginaU-shapeddose-responsefunction.Todetermineifthecisplatindose-responsefunctionforvestibularhaircellsfollowsasimilarpattern,wetreatedvestibularorganotypiccultureswithdosesofcisplatinrangingfrom10to1000μM.Vestibularhaircelllesionsprogressivelyincreasedasthedoseofcisplatinincreasedwithmaximumdamageoccurringaround50–100μM,butthelesionsprogressivelydecreasedathigherdosesresultinginlittlehaircelllossat1000μM.TheU-shapeddoseresponsefunctionforcisplatin-treatedvestibularhaircellsincultureappearstoberegulatedbycoppertransporters,Ctr1,ATP7AandATP7B,thatdose-dependentlyregulatetheuptake,sequestrationandextrusionofcisplatin.

  • 简介:Laser-inducedbreakdownspectroscopy(LIBS)wasexaminedtodetectatracesubstanceadheredontoAlalloysforthesurfaceinspectionofmaterialstobeadhesivelybonded.AsanexampleofSicontamination,siliconeoilwasemployedandsprayedontosubstrateswithacontrolledsurfaceconcentration.LIBSmeasurementsemployingnanosecondUVpulses(λ=266nm)andanoff-axisemissioncollectionsystemwithdifferentdetectingheightswereperformed.Becausesurfacecontaminantsareinvolvedintheplasmaformedbylaserablationofthesubstrates,therelativecontributionofthesurfacecontaminantsandthesubstratestotheplasmaemissioncouldbechangeddependingontheconditionsforplasmaformation.Thelimitofdetection(LOD)wasevaluatedunderseveraldetectingconditionsforinvestigatingthefactorsthataffectedtheLOD.Asignificantfactorwasthestandarddeviationvaluesofsignalintensitiesobtainedforthecleansubstrates.Thisvaluevarieddependingonthemeasurementconditions.FortheAlalloy(A6061),thesmallestLODobtainedwas0.529μg·cm^-2.Furthermore,animprovedLOD(0.299μg·cm^-2)wasobtainedfortheAlalloywithalowerSicontent.

  • 简介:ThepresentstudywasdesignedtodeterminetheeffectsofatraditionalChinesemedicine,calledQishenYiqiDroppingPillonchronichypoxia-inducedmyocardialinjury.ToestablisharatchronichypoxiamodeltobeusedintheevaluationofthetherapeuticeffectsoftheQishenYiqiDroppingPill,Sprague-Dawley(SD)ratswererandomlydividedintothreegroups:thecontrol,model,andtreatmentgroups(n=10pergroup).Theanimalswerehousedinaplexiglasscontainer.Thecontrolanimalswereundernormaloxygenconcentrationandthemodelandtreatmentgroupswereexposedtoairandnitrogenfor5weeks.TheratsinthetreatmentgroupwereorallyadministeredtheQishenYiqiDroppingpill(35mg·kg-1·d-1)for5weeks.Afterthetreatment,thecardiacfunctionandmorphologywereanalyzed,andtheexpressionlevelsofhypoxia-induciblefactor1α(HIF-1α)weredeterminedusingWesternblotting.Ourresultsindicatedthatthecardiacfunctionwasimpaired,cellapoptosiswasenhanced,andHIF-1αexpressionwasup-regulatedinthemodelgroup,comparedtothecontrolgroup.ThesechangeswereamelioratedbythetreatmentwiththeQishenYiqiDroppingPill.Inconclusion,QishenYiqiDroppingpillcanamelioratemyocardialinjuryinducedbychronichypoxia,improvecardiacfunction,anddecreasemyocardialcellapoptosis,whichmayprovideabasisforitsclinicaluseforthetreatmentofchroniccardiovasculardiseases

  • 简介:SeacucumberandcordycepssinensisareusedasbothfoodandtraditionalmedicinesinAsia.Thisstudywascarriedoutinordertoinvestigatethehpyerglycemiceffectofamixtureofseacucumber(Apostichopusjaponicas)andcordycepssinensis(Cordycepsmilitaris)(SCC)indiabeticratandexplorethemechanismunderliningsuchaneffect.Thediabeticmodelratwasinducedwithintraperitonealinjectionofstreptozotocin(STZ).Thediabeticmodelratswererandomlydividedintocontrolgroup(0.9%NaCl),lowdosegroup(300mgSCC·(kgbodyweight)-1)andhighdosegroup(1200mgSCC(kgbodyweight)-1).SodiumchlorideandSCCwereintragastricallyadministeredonceadayfor35d.Changesinfastingserumglucoseandseruminsulincontent,oralglucosetoleranceandliverandmuscleglycogencontentwereroutinelyevaluated.Pancreastissueandβ-cellsofisletswereobservedunderbothopticalandtransmissionelectronicmicroscope,respectively.Theabundanceofglucosemetabolism-relatinggenesingastrocnemiusandepididymaladiposetissuewasdeterminedwitheitherreversetranscriptionPCR(RT-PCR)orwesternblotting.ResultsshowedthatSCCsignificantlydecreasedfastingserumglucosecontent,improvedglucosetoleranceandincreasedseruminsulinandglycogencontent;repairedSTZ-injuredβ-cellsofdiabeticrat,andincreasedtheexpressionofphosphatidylinositol3kinase(PI(3)K),proteinkinaseB(PKB)andglucosetransporter4(Glut4)encodingproteininbothgastrocnemiusandadiposetissue,andGlut4encodinggeneinperipheraltissue.OurfindingsdemonstratedthatSCCexertedananti-hyperglycemiceffectbyrepairingβ-cellsandpromotinginsulin-mediatedsignaltransductionpathwayininsulin-sensitivegastrocnemiusandadiposetissue.

  • 简介:Unlikeatraditionalfiberlaserwithadefinedresonantcavity,arandomfiberlaser(RFL),whoseoperationisbasedondistributedfeedbackandgainviaRayleighscattering(RS)andstimulatedRamanscatteringinalongpassivefiber,hasfundamentalscientificchallengesinpulsingoperationforitsremarkablecavity-freefeature.Forthetimebeing,stablepulsedRFLutilizingapassivemethodhasnotbeenreported.Here,weproposeandexperimentallyrealizethepassivespatiotemporalgain-modulation-inducedstablepulsingoperationofcounterpumpedRFL.Thankstothegoodtemporalstabilityofanemployedpumpingamplifiedspontaneousemissionsourceandthesuperiorityofthispulsegenerationscheme,astableandregularpulsetraincanbeobtained.Furthermore,thepumphysteresisandbistabilityphenomenawiththegenerationofhigh-orderStokeslightispresented,andthedynamicsofpulsingoperationisdiscussedafterthetheoreticalinvestigationofthecounterpumpedRFL.ThisworkextendsourcomprehensionoftemporalpropertyofRFLandpavesaneffectivenovelavenuefortheexplorationofpulsedRFLwithstructuralsimplicity,lowcost,andstableoutput.
