  • 简介:BACKGROUND:Thestellateganglionblock(SGB)playsaprotectiveroleinfocalcerebralischemia/reperfusioninjury.ThehumanSGBcanbesimulatedbytransectionofthecervicalsympathetictrunk(TCST)inrats.OBJECTIVE:ToobservetheeffectsofTCSToninduciblenitricoxidesynthase(iNOS)levelsandcerebralinfarctvolumeinthehippocampusofratswithcerebralischemia/reperfusioninjury,andtoanalyzethemechanismofaction.DESIGN,TIMEANDSETTING:Acompletelyrandomized,controlled,neuropathologicalexperimentwasperformedattheInstituteofNeurologicalDisease,TaiheHospital,YunyangMedicalCollegebetweenMarchandSeptember2006.MATERIALS:Atotalof93Wistarrats,aged17-18weeks,ofeithergender,wereusedforthisstudy.2,3,5-triphenyltetrazoliumchloridewaspurchasedfromChangshaHongyuanBiologicalReagentCompany,China.RabbitiNOSantibodyandgoatanti-rabbitIgGantibodyweretheproductsofWuhanBosterBiologicalReagentCo.,Ltd.,China.METHODS:Tenratswererandomlyselectedforthesham-operatedgroup.Cerebralischemia/reperfusioninjurywasinducedbymiddlecerebralarteryocclusion(MCAO)usingthesuturemethodintheremainingrats.Fortysuccessfulratmodelswererandomlyandequallydividedintothefollowingtwogroups:(1)TCSTgroup:subsequenttoTCST,MCAOwasperformedfor2hours,followedby24hoursreperfusion;(2)modelgroup:ratsunderwentexperimentalproceduressimilartotheTCSTgroup,withtheexceptionofTCST.Ratsinthesham-operatedgroupweresubjectedtoexperimentalproceduressimilartothemodelgroup;however,thethreadwasonlyintroducedtoadepthof10mm.MAINOUTCOMEMEASURES:Following24hoursofreperfusion,functionalneurologicaldeficitswerescored.BraintissuesectionsfromtenratsofeachgroupwereusedtomeasurecerebralinfarctvolumebyTTCstaining.HippocampaltissuesectionsofanadditionaltenratsfromeachgroupwereusedtodetectiNOSlevelsusingthestreptavidin-peroxidaseimmunohistochemis

  • 简介:Objectives:Toinvestigatetheeffectsofadenovirus-mediatedinduciblenitricoxidesynthasegenetransfectiononbladdertransitionalcellcarcinomaT24cells,andtoprovidenovelinsightsandapproachestoclinicaltherapiesagainstbladdertransitionalcellcarcinoma.Methods:Firstly,constructrecombinantadenovirusvectorpAd-iNOSofiNOS,followedbytransfectionofpAd-iNOSintoHECK293packagingcells.Thirdly,harvestrecombinantadenovirusrAd-iNOSafteramplificationandpurificationprocedures.Finally,transfecttherecombinantadenovirusrAd-iNOSintohumanbladdercarcinomaT24cellsandexaminetheeffectofrAd-iNOStransfectiononapoptosisofT24andpossiblemechanism.Results:Asshownbythisstudy,therecombinantadenovirusrAd-iNOSwasconstructedsuccessfully.Thevirustiterwas5.8×108PFU/mLandrecombinantwasverifiedbyPCRanalysis.TransfectionofadenovirusrAd-iNOSintoT24cellscouldinducesecretionofhighNOconcentration,P53proteinexpressionupregulation,aswellaspromotionofT24cellapoptosis.Conclusions:ThetransfectionofhumanbladdercarcinomaT24cellsfromrecombinantadenovirusrAdiNOSwasconfirmedtoinduceintracellulariNOSover-expression,highproductionofNO,up-regulationofintracellularP53expressionandpromotionofcellapoptosis.

  • 简介:Anovelfluorescentprobe9-(4-(1,2-diamine)benzene-N1-phenyl)acridine(DABPA)wassynthesizedforthedetectionofnitricoxide(NO)andcharacterizedbyIR,1H-NMRandEI-MSspectroscopy.Basedonaphotoelectrontransfermechanism,thefluorescenceintensitiesofDABPAwereinvestigatedwiththedifferentconcentrationsofNO.Undertheoptimalexperimentalconditions,thefluorescenceintensityofDABPAhadagoodlinearrelationship(R2=0.9977)withNOconcentrationintherangefrom1×10-7to1.5×10-6mol/Lwithadetectionlimitof1×10-8mol/L.ThecytotoxicityinducedbyDABPAwasevaluatedbytheMTT(3-(4,5-dimethylthiazol-2-yl)-2,5diphenyltetrazoliumbromide)assayforbiologicalapplication.Furthermore,theprobeDABPAhadalsobeensuccessfullyappliedtoreal-timeimageNOproducedinPC12cellsinthepresenceofL-arginine.

  • 简介:Inthepresentstudy,aseriesofnovelnitricoxide-hydrogensulfidereleasingderivativesof(S)-3-n-butylphthalide((S)-NBP)weredesigned,synthesized,andevaluatedaspotentialantiplateletagents.CompoundNOSH-NBP-5displayedthestrongestactivityininhibitingthearachidonicacid(AA)-andadenosinediphosphate(ADP)-inducedplateletaggregationinvitro,with3.8-and7.0-foldmoreeffectivenessthan(S)-NBP,respectively.Furthermore,NOSH-NBP-5couldreleasemoderatelevelsofNOandH2S,whichwouldbebeneficialinimprovingcardiovascularandcerebralcirculation.Moreover,NOSH-NBP-5couldrelease(S)-NBPwhenincubatedwithratbrainhomogenate.Inconclusion,thesefindingsmayprovidenewinsightsintothedevelopmentofnovelantiplateletagentsforthetreatmentofthrombosis-relatedischemicstroke.

  • 简介:Theexfoliationdegreeofgraphiteoxideintographeneoxideplaysanimportantroleinthemassiveproductionmethodofreducedgrapheneoxide.Itissignificanttofindasimpleandfeasiblemethodtoanalyzetheexfoliationdegreeofgraphiteoxide.Inthepresentwork,graphiteoxidewassynthesizedbyamodifiedHummersmethod,andthengrapheneoxidecolloidswereobtainedbyexfoliationofgraphiteoxidedispersedinde-ionizedwater.UV-visiblespectroscopywasusedtocharacterizetheabsorptionofthegrapheneoxidecolloids,andtheconcentrationofgrapheneoxidecolloidsindicatedbyabsorptionareaofUV-visiblespectrawasstudied.Resultsshowthatthereisarelativelystablerelationshipbetweenthem,indicatingthatUVvisiblespectroscopyisapotentialmethodforanalyzingtheexfoliationdegreeofgraphiteoxideintographeneoxide.

  • 简介:OxidemetallurgytechnologyhasbeendevelopedatBaosteelforimprovingtheheataffectedzonetoughnessofsteelplatesforhighheatinputwelding.AfterdeoxidationwithstrongdeoxidizersofMgalloy,thecomplexinclusionscontainingpartsofcompoundsofMgO,Ti2O3,MnO,Al2O3,MnS,CaSandTiNareformed.Thesekindsofinclusionsarebeneficialforpromotingtheformationofintragranularferrite.Aftertwo-electrodevibratoryelectrogasarcweldingwiththeV-typegrooveandahighheatinputof395kJ/cminasinglepass,theformeraustenitegrainisveryfineinsize,withanaveragegrainsizeofonly85μm.Excellentheataffectedzonetoughnessisobtainedforthedevelopedsteelplateswithathicknessupto68mm.

  • 简介:Meltsofleadbismuthgallatecompositionsarehighlycorrosiveandattackoncruciblesofdifferentmaterials.Inthepresentstudy,corrosionbyabaseglass(50PbO-30Bi2O3-20Ga2O3inmolefraction)meltedusingdifferentcruciblesandtheeffectonUV-VISandIRedgeswerestudied.Bymeltingthebaseglassinplatinum/2%rhodium,goldzirconiaandaluminacruciblesshowedlesseffectontheIRedgeandthereforeshiftedtheinfrarededgetolongerwavelength,whereassilicacruciblecontaminatedtheglass,causingaseveredeteriorationintheinfraredandhenceshiftedinfrarededgetomuchshorterwavelength.IntheUV-VISregion,baseglassmeltedinplatinum/2%rhodiumcrucibleshiftedtheedgetothelongestwavelengthwhereassilicacrucibleshiftedtheedgetoshorterwavelength.Thecontaminantsfromgold,zirconiaandaluminacruciblescausedtheUV-VISedgeofthebaseglasstoliebetweenthetwoextremesofPt/2%RhandSiO2crucibles.Theglassesmeltedinabovementionedcrucibleswerealsocharacterizedwithinductivelycoupledplasmaspectroscopy(ICP)analysistomeasurethelevelofcontaminationfromthecrucibles.Dependinguponcrucibleused,thecolorsofglassesobtainedrangedfromredtoyellow.

  • 简介:Aim:Tostudytheeffectofdiabetesmellitusandinsulintreatmentonratpenilenitricoxidesynthasecontent.Methods:MaleWistarratsweredividedatrandomintotwogroups:theControl(n=8)andtheDiabetic(n=17).Diabetesmellituswasinducedbyintraperitonealinjectionofstreptozotocin.Thediabeticanimalswerethenrandomlydividedintotwosubgroups:diabeticratswithoutinsulintreatment(n=7)anddiabeticratswithinsulintreatment(n=10).Theneuronalnitricoxidesynthase(nNOS)inthepenilecorpuscavemosumwereassayedbyimmumohistochemicalstainingwithspecificantibodytonNOSandthenNOS-positivenervefiberswerecountedsemiquantitativelyunderahighpowermicroscope.Results:ThenNOS-positivenervefibresindiabeticratswithtreatmentwashigherthanthatindiabeticratswithouttreatment(P<0.05)andlowerthanthatinthecontrols(P<0.01).ThenNOS-positivenervefibresindiabeticratwithouttreatmentwerealsolowerthanthatinthecontrols(P<0.01).Conclusion:Instreptozotocin-induceddiabeticrats,thenNOScontentinthepenilecorpuscavemosumwassignificantlydecreased.InsulintreatmentatthedoselevelemployedpartiallyrestoresthepenilenNOScontentintheserats.

  • 简介:Industrialapplicationofmicrowave,asaheatingsourceformaterialprocessing,wasreviewed.Thefeasibilityofcarbothermicreductionofzincoxideconcentrate,aswellastheeffectofoperatingparameterswasinvestigated,usingahomestylemicrowaveovenat2.45GHz.Zincoxideconcentratedoesnoteffectivelyabsorbmicrowaveenergy,whileanysourceofcarbon,whichisusedasthereductionagent,absorbsmicrowaveenergyverywell.Inthisrespectcokebreezewasfoundtobethebest,andthus,cokewasusedbothasthereducingagentandtheabsorbentofmicrowaveenergy.Itwasalsofoundthatanyincreaseinthecarboncontentandsize,increasesthereductionrate.Increasingthemicrowavepowerandthesizeofthesamplecouldalsoincreasethereductionrate.Furtherinvestigationshowsthatwhenzincoxideisexposedtothemicrowaveforsometime,therateofthereductionbyconventionalmethodincreases.

  • 简介:Strontium-zinc-oxide(SrZnO)filmsformingthesemiconductorlayersofthin-filmtransistors(TFTs)aredepositedbyusingion-assistedelectronbeamevaporation.Usingstrontium-oxide-dopedsemiconductors,theoff-statecurrentcanbedramaticallyreducedbythreeordersofmagnitude.Thisdramaticimprovementisattributedtotheincorporationofstrontium,whichsuppressescarriergeneration,therebyimprovingtheTFT.Additionally,thepresenceofstrontiuminhibitstheformationofzincoxide(ZnO)withthehexagonalwurtzitephaseandpermitstheformationofanunusualphaseofZnO,thussignificantlychangingthesurfacemorphologyofZnOandeffectivelyreducingthetrapdensityofthechannel.
